I went to Spain in 2004. In 2 weeks, I visited Madrid, Segovia, Toledo, Ronda, Sevilla, Granada and Barcelona.

I loved the contrasts between Madrid, southern Spain, and Barcelona.

I used Madrid as my base for Toledo and Segovia, using public transportation to go to them on day trips, and then picked up the rental car for the trip to southern Spain over to Granada. You can fly cheaply from Granada to Barcelona to save travel time.

For me, I'd say Sevilla and Barcelona were musts, I LOVED Ronda, and Madrid was OK. But it really depends on what you like and what you are looking for.

If you have any more specific questions, I'll try to help.
