where in the US are you flying from, and

We live in a city that doesn't have international routes, so we know we'll have to connect in another US city such as Chicago, Neward, NY, Atlanta, or Boston. The connecting city won't really matter.

what time of year are we talking about

We aren't limited to any particular time of the year for travelling. I've read about the migrations in spring and fall, and the fact that the seasons are reversed on the other side of the equator. just have to drive across the border to get from one to another....if you plan a third week, south africa is a different experience, and in that case you could easily add namibia, which is completely different from everywhere else

So when one talks about a "guided tour," is that with the same guide for multiple areas and in which case the tour company arranges transportation, or does a traveller arrange his own transportation between each destination and then meets up (or not) with a different guide specific to each destination?

Or, can travellers reasonably visit several reserves, guide books in hand and with rental vehicles, and do it all self-guided?
